The mountant of choice for diatom microscopy assays

Diatoms are single-celled algae that live in structures made of glass. They are the only organism on the planet with cell walls composed of transparent, opaline silica. Diatom cell walls are ornamented by intricate and striking patterns of silica which can be used to distinguish the various species. From a microscopy perspective because diatoms have a refractive index very similar to glass to make these very intricate structures visible a mountant with a much higher refractive index is required. Naphrax is now internationally recognised as the mountant of choice.

In solution the refractive index of Naphrax is 1.65. As part of the mounting process the naphrax is heated and the solvent toluene is driven off and the refractive index rises to 1.73.

The microscopy of diatoms benefits significantly from the use of phase contrast and dark ground illumination systems.
